Determination of Infrared Absorbing Cyanine Dye Content for 830nm Digital Plate-Making Materials — High Performance Liquid Chromatography Method
1 Scope
This document describes the method for determining the content of infrared absorbing cyanine dyes used in 830nm digital plate-making materials by high performance liquid chromatography (HPLC).
This document applies to the quality control of benzindole-type cyanine dye products for 830nm digital plate-making materials. The minimum detectable limit for impurities is 0.01%.

3 Terms and Definitions
No terms and definitions are listed in this document.
4 Basic Characteristics
The chemical name of the infrared absorbing cyanine dye product for 830nm digital plate-making materials is 2-[2-[2-chloro-3-[2-(1,3-dihydro-1,1,3-trimethyl-2H-benz[g]indol-2-ylidene)-ethylidene]-1-cyclohexen-1-yl]-vinyl]-1,1,3-trimethyl-1H-benz[g]indolium 4-methylbenzenesulfonate, with CAS No. 134127-48-3. The structural formula is:
Molecular Formula: C₄₇H₄₇ClN₂O₃S
Relative Molecular Mass: 755.41 (based on 2021 International Relative Atomic Masses)
5 Method Summary
Weigh and dissolve the sample. Inject the sample solution into the chromatographic column using a micro-syringe or a fixed-volume injection valve for separation. Detect with a UV-Vis detector at a specified wavelength. Calculate the content of the infrared absorbing cyanine dye for 830nm digital plate-making materials using the area normalization method.
6 Sampling
The sampling method for the infrared absorbing cyanine dye product for 830nm digital plate-making materials shall be carried out according to the provisions of GB/T 6679.
7 Reagents
7.1 Methanol, chromatographic grade.
7.2 Potassium dihydrogen phosphate, analytical grade.
7.3 Water, Grade 1 water as specified in GB/T 6682.
8 Instruments and Equipment
8.1 High performance liquid chromatograph equipped with a variable wavelength UV-Vis detector.
8.2 Analytical balance: Sensitivity 0.1 mg.
8.3 Volumetric flasks: 25 mL.
8.4 Pipettor: 50 μL to 5 mL.
8.5 Filtration apparatus equipped with a 0.22 μm membrane filter.
9 Recommended Chromatographic Conditions
9.1 Chromatographic column: Stainless steel, length 150 mm, inner diameter 4.6 mm, column temperature 35°C, packed with octadecyl chemically bonded phase silica gel, particle size 5 μm.
9.2 Mobile phase: Mix 820 mL of methanol and 180 mL of potassium dihydrogen phosphate aqueous solution (25 mmol/L, adjusted to pH 2.0~2.5 with phosphoric acid). Filter through a 0.22 μm nylon membrane filter under vacuum and degas by ultrasonication for 10 min.
9.3 Flow rate: 0.8 mL/min.
9.4 Injection volume: 20 μL.
9.5 Detector: UV detector, detection wavelength 265 nm.
9.6 Under these chromatographic conditions, a sample chromatogram is provided in Annex A (mobile phase pH 2.2).
10 Test Procedure
10.1 Preparation of Sample Solution
Weigh 25 mg of the sample, accurate to 1 mg. Dissolve and dilute to volume with methanol in a 25 mL volumetric flask. Pipette 2.5 mL of this solution and dilute to volume with the mobile phase in a 25 mL volumetric flask to obtain a test solution with a sample concentration of approximately 0.1 mg/mL. Filter through a 0.22 μm membrane filter before analysis.
NOTE: The specified sample weight in this test is determined jointly by the injection volume, column load, impurity peak detection limit, and detector sensitivity, and may be adjusted appropriately.
10.2 Determination
According to the test conditions recommended in Chapter 9, after the instrument stabilizes, inject the specified volume of the prepared sample solution.
10.3 Calculation
The content of the infrared absorbing cyanine dye for 830nm digital plate-making materials is calculated according to formula (1):
10.4 Precision
Perform two parallel determinations using this method. The arithmetic mean of the two parallel determination results shall be taken as the final result. The relative deviation between the two parallel determination results shall not exceed 0.6%. Data processing shall be carried out according to the rounding rules of GB/T 8170.
11 Test Report
The test report shall include the following content:
a) Information about the sample: batch number, date, time, model of instrument used, etc.;
b) Analytical results;
c) Any unusual phenomena observed during the determination;
d) Any operations not specified in this document or test conditions chosen freely.
