Thermostable α-amylase preparation 1 Scope This standard specifies the terms and definitions, technical requirements, test method, inspection rules, marking, packaging, transportation and storage requirements of thermostable α-amylase preparation. This standard is applicable to the thermostable α-amylase preparation produced by bacillus licheniformis (or other dissociants) via deep fermentation culture and purification. 2 Normative references The following standards contain provisions which, through reference in this standard, constitute provisions of this standard. 5 试验方法 本方法所用的水均为蒸馏水或去离子水，所用试剂在未特殊注明时均为分析纯。 5.1 酶活力的测定 5.1.1 分光光度计法(第一法) 5.1.1.1 原理 α-淀粉酶能将淀粉分子链中的α-1，4葡萄糖苷键随机切断成长短不一的短链糊精、少量麦芽糖和葡萄糖，而使淀粉对碘呈蓝黑色的特异性反应，随淀粉降解蓝色逐渐减褪，渐变成红棕色，其颜色消失的速度与酶活性有关，故可在标准条件下，通过测定反应后溶液的吸光度计算其酶活力。 5.1.1.2 试剂和溶液 a)原碘液 称取碘化钾22.0g溶于约300mL水中，加入碘11.0g，在搅拌下使其溶解，然后移入500mL容量瓶中，用水定容，贮于棕色瓶中备用(每月制备一次)。 b)稀碘液 称取碘化钾20.0g溶于约300mL水中，准确加入原碘液2.00mL，全部移入500mL容量瓶中，用水定容，贮于棕色瓶中备用(须当天配制)。 c)20g/L可溶性淀粉1)溶液 注：1)采用浙江菱湖食品化工联合公司生产的、标有‘专供酶制剂测定'用的字样和批号的“可溶性淀粉”。 称取可溶性淀粉(以绝干计)2.0000g，精确至0.0001g，用水调成浆状物，在搅动下缓缓倾入70mL沸水中，然后，以30mL水分几次冲洗盛淀粉的小烧杯，洗液并入其中，加热煮沸2min直至完全透明，冷却至室温，全部移入100mL容量瓶，用水稀释至刻线。此溶液必须当天配制。 d)磷酸缓冲液(pH＝6.0) 称取磷酸氢二钠(Na2HPO4·12H2O)45.23g、柠檬酸(C6H8O7·H2O)8.07g，用水溶解并定容至1000mL。配好后，用pH计校正。 e)盐酸溶液c(HCl)＝0.1mol/L，按GB 601配制。 5.1.1.3 仪器和设备 分光光度计； 恒温水浴 50℃～100℃精度±0.1℃； 试管 25mm×200mm； 容量瓶； 自动吸管； 秒表。 5.1.1.4 待测酶液的制备 a)液体酶制剂：直接用缓冲液(5.1.1.2d)配制，使最终酶液浓度控制在65～70u/mL范围内。 b)固体酶制剂：称取酶粉1～2g，精确至0.0002g，先用少量磷酸缓冲液溶解，并用玻璃搅棒捣研(或用磁力搅拌器搅拌)，直至固体全部溶解(约10～15min)，将上清液小心倾入容量瓶中，沉渣部分再加入少量缓冲液，如此捣研3～4次，最后全部移入容量瓶中，用缓冲液稀释至刻度(使酶浓度控制在65～70u/mL范围内)，摇匀。通过四层纱布过滤，收集滤液供测定用。 5.1.1.5 测定 a)吸取可溶性淀粉溶液(5.1.1.2c)20.0mL和缓冲液(5.1.1.2.4)5.0mL于试管中，在70℃恒温水浴中预热平衡5min。 b)加入稀释好的待测酶液(5.1.1.4)1.00mL，立即用秒表记录时间，摇匀，准确反应5min。 c)立即用自动吸管吸取反应液b)1.00mL，加到预先盛有0.1mol/L盐酸0.5mL和稀碘液(5.1.1.2b)5.00mL的试管中，摇匀。 d)以0.1mol/L盐酸0.5mL和稀碘液5.00mL的混合液作试剂空白，于660nm波长下，用10mm比色皿迅速测定其吸光度(A)。根据吸光度查附录A(标准的附录)，求得测试酶液的浓度(c)。 5.1.1.6 计算 X＝c×n×16.67 (1) 式中：X——样品的酶活力，u/mL(u/g)； c——测试酶的浓度，u/mL； n——样品的稀释倍数； 16.67——换算常数。 所得结果表示至整数。 5.1.1.7 结果的允许差 平行试验相对误差不得超过2％。 5.1.2 目视比色法(第二法) 5.1.2.1 原理 同5.1.1.1。 5.1.2.2 试剂和溶液 a)原碘液 同5.1.1.2a。 b)稀碘液 同5.1.1.2b。 c)20g/L可溶性淀粉溶液 同5.1.1.2c。 d)磷酸缓冲液(pH＝6.0) 同5.1.1.2d。 e)标准终点色溶液： A液：称取氯化钴(CoCl2·6H2O)0.2439g和重铬酸钾0.4878g，用水溶解定容至500mL。 B液：称取铬黑T(C2OH12N3N·O2S)40mg，用水溶解并定容至100mL。 C液(标准终点色溶液：取A液40mL与B液5.0mL充分混匀，备用。此混合液于冰箱中保存，15天后需要重新配制。 5.1.2.3 仪器和设备 恒温水浴 50～100℃ 精度±0.1℃； 容量瓶； 试管 25mm×200mm； 秒表。 5.1.2.4 分析步骤 待测酶液的制备：按5.1.1.4进行制备，但最终酶液浓度需控制在300～350u/mL范围内。 吸取标准终点色溶液6mL于试管中，作为比色标准。 吸取20g/L可溶性淀粉溶液20mL和pH6.0的磷酸缓冲溶液5mL置于25mm×200mm试管中，在70℃恒温水浴中预热平衡5min，然后加入预先稀释好的酶液0.5mL，立即用秒表记录时间，充分摇匀，当反应接近2min时，就开始不时地用吸管取出反应液1.00mL，加到预先盛有稀碘液(5.1.1.2b)5.00mL的试管中，当试管中反应溶液颜色由紫色渐变为红棕色、恰与标准终点色溶液相同时，即为反应终点，并记录到达终点所需时间(分钟)。 酶反应全部时间控制在2～2.5min之内。 5.1.2.5 计算
